Originally established by the Lamothe family, the estate was taken over by the Despujols family in 1961, leading to its current name. Spanning 7.5 hectares, the estate primarily cultivates Sémillon, Sauvignon Blanc, and Muscadelle grapes.

The vineyard's composition includes 85% Sémillon, 10% Sauvignon Blanc, and 5% Muscadelle. The diverse soil, a blend of sand, gravel, limestone, and clay, contributes to the production of high-quality grapes. With an average vine age of 40 years, some vines are even over 70 years old. At Château Lamothe Despujols, vinification occurs in stainless steel tanks, followed by maturation in both tanks and new French oak barrels. While the wines can be enjoyed young, the finest bottles reach their peak between 9 to 20 years of age.
